Lillies Gold Schiffs Reagent

Lillie’s Cold Schiff Reagent is a colorless to pale yellow staining reagent used in histology and microscopy for the detection of aldehydes produced during tissue staining procedures. It is a modified form of Schiff reagent developed by R. D. Lillie and is widely used in Periodic Acid–Schiff (PAS) staining techniques.

🏭⚗️ Production
Lillie’s Cold Schiff Reagent is prepared by dissolving pararosaniline and sodium metabisulfite in dilute hydrochloric acid at room temperature. The solution is decolorized with activated charcoal and filtered to produce a clear reagent suitable for histochemical applications.

🔬 Properties
Lillie’s Cold Schiff Reagent is a colorless to very pale yellow solution. It reacts with aldehyde groups to produce a characteristic magenta coloration. The reagent is sensitive to light and temperature and is typically stored under refrigerated conditions to maintain stability.

🧪 Applications
In histological and laboratory applications, Lillie’s Cold Schiff Reagent is widely used in Periodic Acid–Schiff (PAS) staining for the detection of glycogen, mucopolysaccharides, basement membranes, and other aldehyde-containing tissue components.
